1.if a combined B/L required is it compulsory more than one transportation

A little bit understanding on phrase: "more than one transportation".
A combined transport B/L is a transport document that covers at least two different modes of transport.
If a credit require a presentation of combined transport document, and if the transport document clearly shows that it covers a shipment from the place of taking in charge or port, airport or place of taking in charge port, airport of loading to the place of final destination mentioned in the credit, UCP600 article 19 is applicable. In such circumstances, the transport document must not indicate that shipment or dispatch has been effected by only one mode of transport, but it may be silent regarding the modes of transport utilized.

two or more modes of transport must be utilised in a combined B/L . Since it is a B/L , one mode of transport must be by sea, and it may be silent regarding the other modes of transport utilised.
P.S. a combined B/L is different from a MTD (multible transport document), as a MTD may not be evidencing a mode of transport by sea.
